Bevacizumab (Avastin), an anti-VEGF antibody is a humanized immunoglobulinG1 (IgG1) monoclonal antibody that inhibits VEGF-dependent angiogenesis. It is one of the most effective drugs widely used for treatment of ocular angiogenesis diseases. Short half-life of this protein in vitreous, necessitates repeated intravitreal injections. In order to improving accessibility and permanency of Avastin during treatment, nanoliposome was employed as an acceptable ophthalmic drug delivery system. In this respect nanoliposome containing Avastin (NLP-AVA)was prepared recruiting thin film evaporation method followed by hydration. Then formulation was characterized through the analysis of its particle size, zeta potential and its morphology. Moreover toxicity and biocompatibility was evaluated by MTT, LDH and hemolysis assays respectively. The temperature stability of bevacizumab (Avastin) during encapsulation was assessed by monitoring significant changes in its secondary structure using fluorescence spectroscopy. The results show that the bevacizumab (Avastin) entrapment into nanoliposome meaningfully kept it safe and stable for ocular drug delivery and it may improve the treatment of ocular neovascular disease.
