Diabetes is a metabolic disorder associated with hyperglycemia and caused by defect in insulin secretion or insensitivity of target organs to insulin. Non-enzymatic glycosylation of proteins and the formation of AGE is one of the factors involved in the pathogenesis of chronic complications of diabetes. Since glycation reactions combined with oxidation reactions disrupt the proper functioning of proteins, the use of plant-based antioxidant compounds can be used as a treatment to prevent these chronic side effects. One of the important genera of family Solanaceae is Hyoscyamus and its most important species are H. niger L., H. muticus L., H. albus L., H. Aureus L., H. reticulates L., H. senecionis Willd. and H. pusillus L. In this study, the effect of methanolic extract of H. senecionis seeds on inhibition of non-enzymatic glycosylation of hemoglobin was investigated in vitro. Then, in order to form AGE, hemoglobin was glycated by glucose in vitro and to evaluate the effect of methanolic extract of H. senecionis seeds on the glycosylation process, the fluorescence intensity of the samples was measured at ۴۴۳ nm .In this experiment, the concentration of the extract required to inhibit ۵۰% of the non-enzymatic glycosylation process of hemoglobin (IC۵۰) was obtained and compared with the required amount of ascorbic acid as a positive control. Seed methanolic extract with IC۵۰ = ۲۵۳.۸۴ μg / ml has anti-diabetic properties compared to ascorbic acid with IC۵۰ = ۸۰,۹۲μg / ml. According to the obtained IC۵۰, the inhibitory effect of methanolic extract of plant seeds is low compared to ascorbic acid.
