Objective(s): This study aimed to determine the collagen type II (COL۲) and SOX۹ expression in interleukin growth factor (IGF-۱)-induced Wharton’s Jelly mesenchymal stem cells (WJMSCs) and the level of chondrogenic markers in co-culture IGF۱-WJMSCs and IL۱β-CHON۰۰۲ as osteoarthritis (OA) cells model. Materials and Methods: WJMSCs were induced with IGF۱ (۷۵, ۱۵۰, and ۳۰۰ ng/ml) to enhance their chondrogenesis capability. The gene expression of SOX۹ and COL۲ was evaluated with quantitative RT-PCR. Furthermore, IGF۱-WJMSCs were co-cultured with IL۱β-CHON۰۰۲ cells in varied ratios (۱:۲, ۱:۱, ۲:۱). Chondrogenic markers ADAMTS۱, ADAMTS۵, MMP۳, MMP۱, and RANKL were measured with ELISA. Results: The IGF۱-WJMSCs had an increased expression of COL۲ and SOX۹. ADAMTS۱, ADAMTS۵, MMP۱, MMP۳, and RANKL levels were decreased in the co-culture IGF۱-WJMSCs and IL۱β-CHON۰۰۲. Conclusion: The IGF۱-induced WJMSCs were capable to enhance chondrogenesis, indicated by increased expression of SOX۹ and COL۲ and decreased expression of ADAMTS۱, ADAMTS۵, MMP۳, MMP۱, and RANKL. These findings can be further used in the osteoarthritis treatment.
