چکیده مقاله:

Backgrounds: microRNAs are involved in important biological processes such as apoptosis,proliferation, differentiation, angiogenesis, and metastasis. Disruption of these processes iscritical to the onset, progression, and treatment of cancer. Many studies showed that miRNAsacted as a complication of tumorigenesis, and these small RNAs were responsible for a variety ofcancers, such as liver cancer. We investigated the effect of miR-۴۲۷۰ inhibitor on metastasis andinvasion in the HepG۲ cell line.Materials and Methods: At first, HepG۲ cells were cultured in RPMI ۱۶۴۰ Medium with ۱۰%FBS and antibiotics at ۳۷°C and co۲ ۵%. Then, HepG۲ cells were treated with variousconcentrations of the specific hsa-miR-۴۲۷۰ inhibitor to achieve the appropriate concentrationsof the specific hsa-miR-۴۲۷۰ inhibitor (۴۰ nM) to inhibit metastasis. Finally, invasions andmetastatic potential of treated cells were investigated relative to control cells using the woundhealing assay.Results: The wound healing assay results indicated that treatment of the HepG۲ cells with miR-۴۲۷۰ inhibitor at ۴۰ nM lead to the decline of in-vitro invasion and metastatic compared tountreated HepG۲ cells as control.Conclusion: our result indicated that miR-۴۲۷۰ inhibitor decreased in-vitro invasion in HCC cellline or HepG۲
