Objectives: Zinc ion as certain essential trace element was reported to be involved in the regulation of bone metabolism. It also helps protein synthesis in osteoblastic cells and plays a role in the preservation of bone mass. In this study, we have aimed to elucidate effects of zinc ion on osteogenic differentiation of rat adipose tissue-derived mesenchymal stem cells (rADSCs). Materials and methods: rADSCs were isolated from rat inguinal adipose tissue. Their viability was assessed by MTT assay. osteoblastic protein expression were performed to assess the effect of zinc ion on osteogenic differentiation of rADSCs. Results: zinc ion supplementation at o.432 μg/ml concentration resulted in inhibition of osteogenesis of rADSCs, along with reduction in expression of osteocalcin (OCN). Conclusions: Our study demonstrated that zinc ion inhibited osteogenic differentiation of rADSCs in vitro.
