One of the heavy metals having wrecking effects on many tissues such as bone marrow is lead. In this research, we consentrated on effects of oral dose of lead acetate II fed by Wistar rat on osteocalcin expression in bone marrow mesenchymal stem cells in osteogenic medium. Eigtheen male Wistar rats were used. six rats were fed orally with lead acetate II (dose of 50 ppm) with libitum feed as first group and six rats were fed with lead acetate II (dose of 100 ppm) along with libitum feed as second group for 2 months but other rats were just fed with distilled water (with-out lead metal) and libitum feed as control group. Next they were killed and mesenchymal stem cells (MSCs) of the femoral bone marrow were isolated, then cultured in an osteogenic medium for 21 days. Finally osteocalcin gene expression of stem cells of two groups compaired to control group were determined by Real time PCR.Real time PCR demonstrated that after feeding rats with lead acetate II, the osteocalcin gene expression of MSCs in osteogenic medium in second group reduced significantly more than first group and first more than control group It seems lead toxicity and its accumulation with doses of 50 and 100 ppm in femoral bone marrow lead to decrease osteocalcin gene expression of MSCs in osteogenic medium
