Shiga toxin producing E. coli is causative agent of both human and animals. Among this, E. coli O157 is highlighted. A sensitive and specific test for rapid diagnosis of causative agents is needed. Three hundred cattle in main slaughterhouse in Shiraz were investigated by conventional plating and multiplex PCR. According to PCR profile, the sensitivity and specificity are 93.3 and 86.6% respectively. Stx2 was distinguished as a dominant gene in E. coli O157 isolates. Simultaneous eae and Stx1 or Stx2 in bacteria was the main risk factor in humans. The bacteria contain Stx2 only, was shown the most cytopathic effect on vero cells. The bacterial excretion was seen in warm season than cold. The relation homology was seen between isolates and foreign serotypes. But phylogenic analysis was revealed, this is a subgroup of gene bank and introduce as an independent serotypes. This is the first report of gene analysis in E. coli O157, but more research was needed to find more reservoirs of ecosystem of organisms
