Lipoprotein lipase gene polymorphisms as risk factors for stroke: a computational and meta-analysis

Objective(s): Stroke is the most common neurological disorder and genetic susceptibility has an important role in its etiology. Polymorphism in several genes such as lipoprotein lipase (LPL) is propounded as a risk for stroke. This meta-analysis investigated the association of rs۲۸۵ and rs۳۲۰ LPL polymorphism with stroke risk. Materials and Methods: We searched PubMed, Clarivate Analytics Web of Science, Google Scholar, and Science Direct databases for appropriate studies. The odds ratios (ORs) with ۹۵% confidence intervals (CIs) were calculated to estimate the strength of this association. Also, the effects of four common polymorphisms (rs۲۶۸, rs۲۸۵, rs۳۲۰, and rs۳۲۸) on the molecular aspects of LPL were evaluated by in silico tools. Five studies were included in meta-analysis after screening. Results: Our data indicated that rs۳۲۰ significantly decreased the risk of stroke (G vs. T: OR= ۰.۶۴, ۹۵%CI=۰.۵۴-۰.۷۶; GG vs. TT: OR=۰.۴۷, ۹۵%CI=۰.۲۹-۰.۷۵; TG vs. TT: OR=۰.۶۵, ۹۵%CI=۰.۵۳-۰.۸۰; TG+GG vs. TT: OR=۰.۶۲, ۹۵%CI=۰.۵۱-۰.۷۵; GG vs. TT+TG: OR=۰.۵۱, ۹۵%CI=۰.۳۲-۰.۸۲). Moreover, a significant association between rs۲۸۵ and diminution of stroke risk was seen (P- vs. P+: OR=۰.۷۲, ۹۵%CI=۰.۵۸-۰.۹۱; P-P- vs. P+P+: OR=۰.۵۰, ۹۵%CI=۰.۳۱-۰.۸۲; P+P-+P-P- vs. P+P+: OR=۰.۷۲, ۹۵%CI=۰.۵۳-۰.۹۶; P-P- vs. P+P++P+P-: OR=۰.۵۸۱, ۹۵%CI=۰.۳۶۹-۰.۹۱۶). Also, the same results were observed after stratifying, without any publication bias (PEgger>۰.۰۵). Furthermore, computational analysis revealed that rs۲۶۸ and rs۳۲۸ may affect the protein structure (prediction: non-neutral; score=۱۹; expected accuracy=۵۹%) while rs۳۲۰ could affect the RNA structure (distance=۰.۲۲۶۴, P-value=۰.۰۵۳۴; PConclusion: This meta-analysis indicated that risk of stroke was decreased in rs۳۲۰ and rs۲۸۵ polymorphisms in the LPL gene.
