چکیده مقاله:

Objective: The aim of present study was to investigate susceptibility of PPD plus health care workers to Coronavirus disease ۲۰۱۹ (Covid-۱۹). For this reason,single nucleotide polymorphism (SNP) of INF-γ R۱ at position -۵۶ and INF-γ gene at position +۲۱۰۹ assessed PPD plus group before and after covid-۱۹ pandemic(۲۰۱۷-۲۰۱۸, ۲۰۲۰-۲۰۲۱). Methods: The selected study cases (N=۱۰۰) had PPD positivity >۱۵ (۱۶-۲۰) and duration of their working in TB unite was from minimum ۵ to maximum ۱۰ years. ۵۰ DNA sample were collected from gene bank of TB instituted that belong to TB stuff (group A). The other ۵۰ samples were collected in the same unite during covid-۱۹ pandemic (group B). The SNP in INF-γ R۱ and INF-γ gene at position - ۵۶, +۲۱۰۹ performed using specific primer by and the PCR products -۵۶ (۳۶۶ bp) and +۲۱۰۹ (۶۷۰ bp) digested using restriction enzyme Bts I and Fau I, respectively. Statistical analyses were used to obtain the frequency of alleles among all static Cassese. The confidence intervals (CI) and t test were calculated using SPSS and Graph Pad Prism software.Result: In overall, the most frequent genotype in group A was AA (۴۱/۵۰; ۸۲%) and in group B was ۷۶% (۳۸/۵۰) in position +۲۱۰۹ and frequency allele T wasdominate in two group. The frequency of TC+CC genotype [(OR= ۵.۴۴, ۹۵% CI, ۲.۳۱-۱۲.۸۰ and p=۰.۰۰۰)] in group B was more than group A. the frequentgenotype of TT at position -۵۶ was (۳۵/۵۰; ۷۰%) in group A and group B (۱۵/۵۰; ۳۰%) and allele C of IFN-γ gene at position +۲۱۰۹ (OR= ۴.۴۷, ۹۵% CI, ۱.۷۴-۱۱.۴۳and p=۰.۰۰۲) was significant more than in group B. although the dominate allele in group A and B was allele A at INF-γ R۱ position -۵۶ and allele T in INF-γ gene at position ۲۱۰۹.
