چکیده مقاله:

Objective: Delay in wound healing gives severe health andeconomic challenges to patients and public health especiallyafter surgeries. MSCs (mesenchymal s tem cells) as multipotentnon-hematopoietic progenitors can play a crucial role in tissueregeneration and contribute to wound healing by growth factorssecretion.Materials and Methods: In this s tudy, ۲۰ male Wis tar rats(۲۵۰ ± ۲۰ g) after es tablishing a square wound on the dorsalside were randomly divided into two groups: I. control group,II. hUC-MSCs CM (human umbilical cord mesenchymal s temcells conditioned medium) treatment group. In the treatmentgroup, ۱ cc of Wharton´s jelly-derived hUC-MSCs conditionedmedium was injected intradermally into the peripheral area ofthe wound. The rate of wound healing was measured on the ۰th,۵th, ۱۰th and ۱۴th days and the wound area was marked. Allanimals were euthanized ۱۴ days after the injection and the skinsamples were removed and frozen immediately for complementaryassessments. The expression of TGFβ-۱ and TGFβ-۲(transforming growth factor) were assayed by real-time polymerasechain reaction (PCR).Results: The results showed that the expression of TGFβ-۱ wassignificantly increased in comparison with the control group(P value <۰.۰۵). Although the expression of TGFβ-۲ was increasedin comparison to the control group but it was not significant.Also, the assessment of the healing of the wound areashowed that the treatment was remarkably effective in woundshealing (P value <۰.۰۵).Conclusion: Human umbilical cord mesenchymal s tem cellsconditioned medium have an effective contribution in thewound amelioration and it can increase the level of TGFβ-۱expression.
