چکیده مقاله:

Background: In this study, the efficacy of riboflavin (VB۲) in preventing uterus As۲O۳ damage was investigated for the first time in the literature. Methods: The rats received ۴۰ μg LHRHa for estrus synchronization. ۴۸ pregnant Wistar rats were included in the study. Four groups were formed with ۷ rats in each group: Sham, ۱.۵ mg arsenic trioxide (As۲O۳/L) alone or in combination with VB۲ (۲۰ and ۴۰ mg/L) in drinking water (for ۲۱ days continuously). Moreover, similar to maternal generation treatment the F۱-female generation was arranged (for ۳۵ days continuously until puberty). Results: Based on the results, As۲O۳ reduced body weight and feed intake (P < ۰.۰۵). Furthermore, the serum malondialdehyde levels in the As۲O۳ group were significantly higher than that of the control group (P < ۰.۰۵). At the same time, the total antioxidative status and the activities of glutathione peroxidase, superoxide dismutase, and catalase were reduced (P < ۰.۰۵). Meanwhile, As۲O۳ remarkably increased the inflammatory markers production [interleukin ۶ and C-reactive protein] (P < ۰.۰۵). As۲O۳ administration induced uterus apoptosis-related genes by upregulating caspase-۳, iNOS, and Bax genes and downregulating Bcl-۲ gene of pubertal F۱-female rats (P < ۰.۰۵). Conclusion: Our observation indicated that VB۲ therapy is potentially an effective strategy to modify the detrimental effects of As۲O۳ in pubertal F۱-female rats via suppresses oxidative damages.
