Up-regulation of LINC۰۲۵۴۲ reduced thesurvival rate of colorectal cancer patients: high-throughput data analysis in a systems biology approach

Background: Colorectal cancer (CRC) is the third most prevalentmalignant tumor affecting humans worldwide, as well as asignificant cause of mortality. Many deaths could be avoided ifprecancerous adenomas or early-stage cancers could be identifiedand treated prior to the development of more advanced malignancy.This experiment aimed to recognize the differentiallyexpressed genes in the carcinogenesis and progression of CRCvia bioinformatics analysis.Materials and Methods: The GSE۱۱۰۷۱۵ (۶ tumor and ۶ controlsamples) was selected from GEO datasets and analyzedusing GEO۲R. Adjusted P-value < ۰.۰۵ and |logFC|> ۲ wereconsidered as statistical significances. Functional and pathwayenrichment analysis was performed for DEGs using DAVIDdatabase. The analysis of the DEGs pathways was done in theKEGG online database. lncRNA interactions analysis was performedusing lncRRIsearch.Results: Based on GEO analysis LINC۰۲۵۴۲ (logFC:۲.۴۵۶۷۷۴۸۳۹, adj. P. Val: ۰.۰۰۰۵۱۳۰۳۹) and LINC۰۱۱۳۲ (log-FC: ۲.۳۶۱۸۷۰۵۷۱, adj. p. Val: ۰.۰۰۱۶۶۶۵۳۷) were chosen asdifferentially expressed genes, which could considered as thenovel diagnostic biomarkers. GEPIA۲ and ENCORI data analysisvalidates the microarray analysis Results: High expressionof LINC۰۲۵۴۲ has a significant correlation with lower survivalrate of CRC patients. LINC۰۲۵۴۲ has significant interactionwith IGF۲ gene (۱۸ interactions). LINC۰۱۱۳۲ has significantinteraction with MUC۲ gene (۲ interactions). IGF۱ and MUC۲has a significant role in cancer signaling pathway. FOXG۱ andNEUROD۱ (neuronal differentiation) have significant interactionswith LINC۰۲۵۴۲ (۱۸,۱۷interaction). as well as FOXG۱has intraction with NEUROD۱. FOXG۱ gene has an importantrole in the FOXO pathway.Conclusion: To sum up, this study could be suggested novelinteraction between lncRNA and mRNA (LINC۰۲۵۴۲ -IGF۲-FOXG۱-NEUROD۱-LINC ۰۱۱۳۲-MUC۲) for the candidatediagnostic and prognostic markers associated with CRC by bioinformaticsanalysis. Thus, this result may be considered as apotential curative purpose for CRC patients.
