Background: Colorectal cancer (CRC) is a highly heterogeneouscancer, and the survival rate of colorectal cancer patientsremains poor. Here, we aimed to identify the prognosis-relatedgenes and genes associated with drug resistance in CRC.Materials and Methods: RNA-Seq data were downloadedfrom the TCGA database, and differential expression analysiswas performed between tumor and normal samples. Using univariateand multivariate COX regression analysis and MCODEclustering module, we identified the key prognostic genes inCRC based on the Protein-protein interaction (PPI) network.Survival curves of the high-risk group and low-risk group weregenerated using the Kaplan-Meier method. Quantitative realtimePCR was used to confirm the results of the RNA-Seq datasets.Results: Seven hub genes, including HOXC۶, HOXC۱۳,HOXC۸, and TBX-۱۵ associated with poor prognosis with ahazard ratio (HR) >۱, and SHDB, COX۵A, and UQCRC۱ associatedwith good prognosis with HR<۱ were selected as independentprognostic factors. Based on the risk score model, thenumber of dead patients in the high-risk group was significantlyhigher than in the low-risk group. Finally, the level of somegenes was evaluated in ۳۰ colorectal cancer tissues.Conclusion: Our results indicated that the level of some geneslike TBX-۱۵ and SDHB was up and down-regulated, respectively,in CRC and abnormal gene expression related to poorprognosis and low survival rate of patients.
