miR-۹۹a-۳p and lncRNA XIST modulatesthe interleukin signaling pathway through the interactionwith LCN۲ in colorectal cancer patients

Background: A lipocalin family protein is encoded by thegene LCN۲. Members of these family transport lipids, steroidhormones, and retinoids, which are all tiny hydrophobic compounds.This gene produces a protein called neutrophil gelatinase-associated lipocalin, which limits bacterial developmentby securing iron-containing siderophores. This protein aids ininnate immunity. This protein is a preliminary indicator of acutekidney damage seen in both urine and blood. In this research,we performed a systems biology approach to investigate the interactions,expression patterns, and signaling pathways in colorectalcancer patients.Materials and Methods: GSE۴۱۰۷ microarray datasets wereanalyzed using R Studio (۴.۲.۳). Using the GEOquery package,raw microarray data was downloaded and stored in R Studio.Limma package performed the statistical analysis. UsingSTRING, lncRRIsearch, and miRWalk online software, protein-protein interaction, lncRNA interaction, and miRNA interactionanalyses were performed, respectively. Pathway enrichmentand gene ontology (GO) analyses were performed usingthe enrichr database.Results: LCN۲ has a significantly high expression in colorectalcancer samples (logFC: ۶.۰۲۷, adj. P. Val < ۰.۰۰۰۱). LCN۲has remarkable interaction with MC۴R, MMP۹, SAA۱, and IL۶proteins. lncRNA XIST regulates the expression level of LCN۲(number of interactions: ۷). LCN۲ has a significant interactionwith miR-۹۹a-۳p (position: ۳’UTR, Score: ۱). LCN۲ regulatesInterleukin-۴ And Interleukin-۱۳ Signaling pathway (adjustedp-value: ۰.۰۱۳۳۷). Based on GO analysis, LCN۲ protein involvesin the iron coordination entity transport process via ironion binding molecular function in specific granule lumen.Conclusion: miR-۹۹a-۳p modulates Interleukin-۴ And Interleukin-۱۳ Signaling pathway through the suppression of theexpression level of LCN۲ mRNA (۳’UTR interaction). LCN۲is a potential oncogene (up-regulated mRNA) and diagnosticbiomarker of colorectal cancer. Changes in the expression levelof lncRNA XIST and miR-۹۹a-۳p and disturbance in the inter action of mentioned RNAs with LCN۲ might increase the riskof gastric cancer
