Altered expression of lncRNA MIAT/miR-۱۷-۵p/ miR-۳۰۲/ miR-۱۰۶b/ miR-۳۶۳ is associated with gliomachemo-resistance

Introduction: Myocardial infarction-associated transcript(MIAT) is a long non-coding RNA (lncRNA) with altered expressionin different diseases and malignancies. An increasingnumber of recent studies have documented that lncRNAs wereassociated with the development of drug resistance to standardchemotherapy regimen in glioma. The involvement of lncRNAMIAT in drug resistance associated with glioma is not clear.Therefore, in this study we have evaluated the role of lncRNAMIAT in glioma and its suppression effects on multidrug resistance(MDR) and its associated miRNAs.Methods: In this study, CCLE (Broad Institute Cancer CellLine Encyclopedia) data were used to evaluate the associationof lncRNA MIAT expression with MDR and sensitivity. Then,lncRNA MIAT was knocked down in two glioma cell lines,U۸۷-MG and A۱۷۲, using RNA interference (RNAi) strategy.Following MIAT down regulation, the expression level of MDRassociated miRNAs was also evaluated using quantitative Real-Time PCR method.Results: The data showed that the increased expression oflncRNA MIAT was associated with resistance to Paclitaxel,PLX۴۷۲۰, and Topotecan chemotherapy drugs. In contrast,down regulation of lncRNA MIAT in two glioma cell lines,U۸۷-MG and A۱۷۲, resulted in deregulation of associated miRNAsincluding miR-۱۷-۵p, miR-۳۰۲, miR-۱۰۶b and miR-۳۶۳.Conclusion: Altogether, our data suggested that MIAT couldinvolve in MDR by targeting miRNAs in glioma, althoughsome more cellular and molecular investigations are also neededto verify these results.
