Antiproliferative activity of Lactobacillus buchneri probiotic bacteria inHT-۲۹ colon cancer cells

Based on GLOBOCAN ۲۰۱۸ data, colorectal cancer (CRC) is the third most deadly andfourth most detected cancer worldwide. However, numerous modern medical methods areavailable for CRC treatment, the survival rates are poor with some unfavorable treatmentrelatedconsequences, which affects the quality of life. The current experimental evidencesupported that probiotics and their metabolites keep the CRC patients safe from treatmentrelatedside effects. Therefore, the current study aimed to evaluate the effect of cell freesupernatant (CFS) prepared from Lactobacillus buchneri isolated from traditional dairyproducts and to investigate its cytotoxic activity and apoptosis induction characteristics onHT-۲۹ colon cancer cell line. MTT assay and Annexin V-FITC analysis were performed toevaluate the induction of apoptosis in HT-۲۹ treated cells. The CFS of L. buchneri preventedthe HT-۲۹ cancer cells growth in a dose- and time-dependent manner, which it showed thehighest anticancer activity at ۲۰۰ μg/mL after ۷۲ h. The apoptotic activity of CFS was alsoconfirmed by flow cytometry results, which showed the highest incidence of apoptosis in HT-۲۹ cancer cells treated with IC۵۰ concentration of CFC after ۷۲ h. The cell cycle analysis alsoreported that the treatment of HT-۲۹ cancer cells with the This investigation demonstratesthat CFS of L. buchneri obtained from Iranian traditional yogurt can protect patients fromCRC development by inducing apoptosis in cancer cells, which can be employed as aprotective anticancer agent in controlling CRC.
