Schiff-base ligands have many biological properties, such as anti-cancer, anti-bacterial and these compounds with their reactive azomethine linkage illustration significant inhibitory effects againstmalignant cells. Copper due to its ability to binding and positive potential of redox is a suitable agent for medical applications. Due to the presence of heterocyclic bases, Cu(II) complexes can be bound tothe DNA and leads to the stopping of the cell cycle and the induction of cell death. Various investigations show that the copper complex has antineoplastic potency against tumor cells. Therefore, in this study, the effect of copper complex toxicity on breast cancer cells has been investigated. In this study, the MTT test was used to evaluate the cytotoxic effects of Schiff-base on breast cancer cells(Mcf7). For these purposes, the cells were seeded and treated with various doses of Cu(II) for 24, 48 and 72 h. Then the capacity of cell inhibition was evaluated by adding of MTT solution (5mg/ml) for4h in dark. In the following, the MTT solution was emptied and 100 μl DMSO was added to each of the wells and its absorbance was measured at 570 nm. The viability of cells was calculated through thefollowing formula OD sample/OD control×100 In addition, morphological changes of the cells were evaluated under an introspective microscope. The obtained results of this investigation confirm the cytotoxic capacity of Schiff-base against breast cancer cells with IC50 about 62.5μg/ml in dose and time-dependently. So that, with increasing time and dose of the treatment, increased the inhibition of the cells and the survival rate is reduced. The results indicate that the survival rate decreased from about 83% (in 30μg/ml) to 11% (in 500μg/ml). The morphological observations showed that deformation of cells has occurred from a multifaceted to spherical. Also, the fragmentation of the core, the wrinkling of the cell was observed. Confirmation of the cytotoxic effect of Schiff-base on cancer cells introduces this substance as an appropriate candidate for use in cancer therapy
