A novel Nano-Carrier of Hydrocortisone 17-Butyrate Based on Ethosome; Synthesis and Characterization

In this research, nano-carriers based on ethosomes were synthesized using lecithin, ethanol and cholesterol with different ratio by hot method. In addition, hydrocortisone 17-butyrate as anti-inflammatory drug was loaded onto ethosomes simultaneously. Synthesized hydrocortisone 17-butyrate loaded- ethosome was characterized using several techniques such as, scanning electron microscopy (SEM), transmittance electron microscopy (TEM), dynamic light scattering method (DLS), zeta potential and turbidimetery. The results of analysis indicated that the particle size of ehosomes was 244 nm in optimize synthesis with high stability after 30 days. The zeta potential result showed a negative value for all of formulation about -4.5 which this value was acceptable in synthesized ethosomes. Additionally, entrapment efficiency of hydrocortisone 17-butyrate onto ethosoms was investigated by a high performance liquid chromatography (HPLC) approach as a reliable method, which the area under the curve each of the samples showed values from 37.15 to 62.92%. The stability of the particle size was measured as a stability criterion for synthesized nanostructures in several times using DLS technique. At result, nano-carriers was synthesized as one pot with a suitable size and high stability to deliver of hydrocortisone 17-butyrate through skin
