چکیده مقاله:

Genosensors have been dev eloped rec ently because of their simplicity, low cost, high sensitivity, rapid analysis and compatibil ity with micro fabricati on technolo gy [۱]. Sequence-specific detection of DNA pr ovides the basis for dete ction of a w ide variety of infectious and inherent disease. lectrochemical hybridiz ation biose nsors for the detection of DNA sequences reduce the assay time and simplify medical analysis [۲]. The gene mutati on takes pla ce in aroun d ۶۰% in all humans' lifetimes ۳. Therefore, introduction of new met hods for mutation detection is very important. Different m ethods are used to an alysis the m utation status of indivi dual tumours. The tumo ur suppressor p۵۳ has been signified in a expanded num ber of biological processes, includin g cell cycle arrest, senescence, apoptosis, autoph agy, metab olism, and a ging. Activation of p۵۳ in response to oncogenic stress deletes nascent tumour cells by apopt osis or sene scence. Improved study ing of the p۵۳ pathway may lead t o better diag nosis and tr eatment of cancer in the future [۴].Herei n, an electrochemical DNA sensor based on a “ sandwich” detection st rategy is rep orted which involves D NA biotiny lated capture probe im mobilized on streptavidin coated nano-magne tic beads using screen printed electrodes (SPEs) and microfluidic based platfo rm or gravi chip. The no velty of thi s work is the combinatio n of a sensitive electroc hemical platform and a proper microfluidic with a simple and effective enzyme sig nal amplifi cation technology (Enzyme-linked Immunosorbent Assays, ELISA) fo r detection of target D NA sequence and single nucleotide mutation in p۵۳ tumor suppressor g ene corresponding oligonucleotide.
