Severe acute respiratory syndrome coronavirus ۲ (SARS-CoV-۲) started a pandemic in the world that resulted in the deaths of more than ۶.۵ million people. Additionally, the efficien-cy of vaccines is decreasing daily due to new variants that can be harmful during the initia-tion of a pandemic. The RNA interference (RNAi) method is a mechanism to stop the tran-scription and expression of target proteins, and RNAi has high efficiency in blocking viral genes. Small interfering RNAs (siRNAs) might be a promising tool for curing viral infections. SARS-CoV-۲ is a respiratory virus that can infect lung cells and transfer easily to another cell. Therefore, the delivery system of designed siRNAs is very important to target lung cells, as mutations in viruses can cause resistance to them. The target genes of siRNA, such as spike protein (S), membrane protein (M), and others, were targeted in past research that demon-strated a new way of anti-viral therapy against respiratory viruses. This work showed that the siRNA treatment for respiratory viruses can be a major way to prevent the infection. In this study, new ideas for targeting the genome of SARS-CoV-۲ based on recent research are suggested that might help in controlling the virus and explain the development of siRNA in an ancestral family of coronaviruses.
