Ketotifen protects mice against gentamicin-induced oxidative stress and nephrotoxicity

چکیده مقاله:

Introduction: Gentamicin is one of the most important and effective aminoglycoside antibiotics in treatmentresistantinfections, whose frequent use leads to severe kidney toxicity. Ketotifen fumarate is a first-generationantihistamine drug and a strong stabilizer of mast cells, which has antioxidant, anti-inflammatory and antiapoptoticproperties in various models of kidney damage. The aim of this study was to investigate the protectiveeffect of ketotifen on gentamicin-induced nephrotoxicity in mice.Methods: In this study, the ameliorative effects of KTF (۲ and ۳ mg/kg, ip) on GEN (۱۰۰ mg/kg, ip) model ofkidney damage were evaluated in male mice. NMRI mice were randomly divided into ۶ groups with ۶ mice ineach group. KTF and GEN were given daily for ۷ days. On the eighth day, the animals were anesthetized, andtheir kidney tissue were removed.Results: The results indicated that KTF can ameliorate GEN-induced the significant increase in TBARS, TNF-α, and NO and decrease in the amount of total thiol, catalase, glutathione peroxidase, superoxide dismutase. Thehistopathological results confirmed indicated findings.Conclusion: The findings of this study demonstrated that ketotifen is reasonably effective in nephrotoxicitygentamicin-induced in mice
